Epitope mapping

Epitope mapping involves the precise definition of the binding site of an antibody to its target protein. Detailed knowledge of the interactions sites on single amino acid level is crucial for understanding the mode of action and biology of an antibody. FDA and EMEA guidelines require such specific binding site information to be included in regulatory dossiers. Also, exact epitope definition is of paramount importance for generating IP protection for novel antibodies.
Pepscan's CLIPS Epitope Mapping Technology allows for the characterization of an epitope at the level of individual amino acids and can be applied to linear, conformational, discontinuous, and complex epitopes involving dimeric or multimeric protein complexes.

Advantages:

  • Provides epitope information at the level of single amino acids
  • Applicable for linear, conformational and discontinuous epitopes
  • Applicable for complex epitopes on dimers, trimers and multimers
  • Side-by-side epitope comparison of different target proteins
  • Benchmarking against known antibodies
  • Screening of FAB's, scFv, nanobodies, or other antibody fragments
  • Identification of new distinct epitopes through comparison with known antibodies
  • No prior structural information on target required
  • No need for recombinant work or crystallization studies
  • Unbiased screening of up to 50 antibodies for the same peptide array

Requirements:

  • Linear sequence of target protein
  • 100 µg antibody/sample

Deliverables:

  • Report including heat-maps, box-plots and 3D visualization of all epitopes
  • Detailed information on epitope for regulatory purposes
  • Detailed epitope information for new IP generation

Antibody Fingerprinting

Pepscan's CLIPS Epitope Mapping Technology allows for the classification of sets of antibodies with respect to the different epitopes recognized. The technology can be used for the benchmarking of lead antibodies against those already available on the market, or for the investigation of new antibodies with respect to IP generation or infringement.

Advantages:

  • Early lead antibody classification
  • Determination of most suitable antibody with respect to epitope and target
  • Benchmarking against known antibodies
  • Information useful for new IP generation or infringement protection

Requirements:

  • Linear sequence of target protein
  • 100 µg antibody/sample

Deliverables:

  • Report including heat-maps, box-plots and 3D visualization of all epitopes
  • Detailed information on epitope for regulatory purposes
  • Detailed epitope information for new IP generation

Biosimilar Validation

Pepscan's CLIPS Epitope Mapping Technology allows for the comparison of binding profiles up to the level of individual amino acids of the epitope. For a novel biosimilar the similarity of the binding profiles between the biosimilar and the originator antibody can be used to validate the biosimilar.

Requirements:

  • 100 µg Biosimilar and 100 µg known (blockbuster) antibody

Deliverables:

  • Report including heat-maps, box-plots and 3D visualization of all epitopes
  • Detailed information on epitope for regulatory purposes
  • Detailed epitope information for new IP generation
