Ligand-Binding Site Analysis

Pepscan's CLIPS Epitope Mapping Technology is not restricted to antibodies but can also be used to identify ligand binding sites. Instead of screening the peptide arrays with an antibody a tagged ligand is used. If a tagged ligand is not available, binding can also be determined through the use of a secondary antibody that binds the ligand.

Advantages:

  • Provides binding site information at the level of single amino acids
  • Applicable to both peptide and hapten ligands
  • Multiple ligands can be compared side by side
  • No prior structural information on target required
  • No need for recombinant work or crystallization studies

Examples of Applications:

  • Ligand binding sites of G-protein coupled receptors
  • Toxin binding sites on ion-channels
  • Ligand binding sites on enzymes

Requirements:

  • Linear sequence of target protein
  • 100 µg of a tagged ligand or 100 µg of the ligand plus an anti-ligand antibody

Deliverables:

  • Detailed information on ligand binding site
  • Information useful for lead development and optimization of peptide therapeutics

Protein-Protein Interaction

Pepscan's CLIPS Epitope Mapping Technology is not restricted to antibodies but can also be used to resolve known interaction sites of proteins in detail. Instead of screening the peptide arrays with an antibody a tagged protein is used. If a tagged protein is not available, binding can also be determined through the use of a secondary antibody that binds the protein.

Advantages:

  • Provides binding site information at the level of single amino acids
  • Suitable for all protein-protein interactions
  • No prior structural information on target required
  • No need for recombinant work or crystallization studies

Examples of Applications:

  • Hormone – receptor interactions
  • Cell matrix interacting proteins
  • Enzyme – substrate interactions
  • Identification of new substrates for enzyme active sites

Requirements:

  • Linear sequence of only one of the interaction target protein
  • 100 µg of a tagged protein or 100 µg of protein plus an anti-protein antibody

Deliverables:

  • Detailed information on protein-protein interactions
  • Information for identification or optimization of new protein therapeutics
